This route provides service on Queen Street between Roncesvalles Avenue and Neville Park Boulevard. It also provides service on The Queensway between Roncesvalles Avenue and Humber Loop, and Lake Shore Boulevard West between Humber Loop and Long Branch Loop. The eastern terminal is Neville Park Loop and the western terminal is Long Branch Loop for its one branch, 301.

- June 23, 2019 - Route made accessible using low-floor streetcars.
- January 3, 2021 - Route partially operated by buses due to construction at King-Queen-Queensway-Roncesvalles intersection, TTC trackwork, and overhead upgrades. Streetcars operate between Neville Park Loop and York Street and buses using the 301L branch designation will operate between Jarvis Street and Long Branch Loop.
- April 5, 2021 - During Phase 1 of the KQQR construction requiring the closure of the Queen Street West portion of the intersection, temporary 301L buses will operate between Long Branch Loop and Jarvis Street, and divert both ways via Dufferin Street and King Street West. Streetcar service now terminates in the west at McCaul Loop instead of operating an on-street loop via York Street.
