This route primarily services Yonge Street between Queens Quay and Steeles Avenue. Its southern terminal is Queens Quay and the northern terminal is Steeles Avenue East for its one branch, 320.

History
- August 1, 2004 - Early morning trips now service the Eglinton Station bus terminal.
- September 7, 2004 - Branch 320B to Queen's Park is introduced providing new overnight service to the hospitals and the Entertainment District. Buses operate via west on Front Street West and north on University Avenue looping via Queen's Park Crescent.
- November 26, 2006 - Branch 320B is discontinued due to low ridership.
- February 18, 2007 - Route is made accessible with low-floor buses.
- June 2, 2011 - Due to lane restrictions on Front Street West stemming from construction on the Union Station expansion project, the south-end looping is temporarily changed to operate via west on Wellington Street West, south on Bay Street, east on Queens Quay West, and north on Yonge Street to route.
- January 3, 2016 - The temporary route change at the south-end looping is made permanent. Buses will no longer operate on Front Street West.
- March 28, 2021 - Some runs on this route now operate out of the brand new McNicoll Division.
- February 16, 2025 - Route is now scheduled to use articulated buses on some runs daily.
