Route details

This route services Spadina Avenue between Bloor Street West and Queens Quay West. It also provides service on Queens Quay West between Lower Spadina Avenue and Bay Street. There is one northern terminal, which is Spadina Station. Southern terminals include King Street West for 510C, Queens Quay & Spadina for 510B, and Union Station for 510A. The 510C short turn branch to King Street West only operates whenever a short turn is required.

History
- June 22, 1990 - 604 Harbourfront begins service running from Union Station to Spadina Avenue.
- July 27, 1997 - Route extended from Queens Quay to Spadina Station. Route becomes 510 Spadina.
- June 17, 2012 - Route is replaced by buses as reconstruction of the tracks and passenger islands is carried out. Modifications are required to meet accessibility standards for the new LRVs that are to enter service in 2014.
- July 5, 2012 - The Shuffle Demons perform their 1986 single "Spadina Bus" along Spadina Avenue on TTC 2252, a 1982 GM New Look.
- August 31, 2014 - Debut of the Bombardier Flexity
streetcars on the route.
- May 10, 2015 - Route is added to the Ten Minute Network with existing service levels over the entire route between Spadina and Union Stations operating at intervals of ten minutes or better whenever the subway is open.
- January 3, 2016 - Route declared fully accessible.
- May 9, 2021 - Weekday service is adjusted to operate on the 510B branch only.
- September 7, 2021 - Weekday branch 510A service is restored.
- June 23, 2024 - Due to overhead replacement along the Spadina right-of-way and construction within the Spadina Station streetcar loop, the route is converted to bus operations serving curbside stops. Replacement buses will operate between Spadina Station and Queens Quay & Spadina loop only looping via Lake Shore Boulevard West, Dan Leckie Way, and Queens Quay West.
- July 8, 2024 - As a result of excessive afternoon rush hour traffic along Spadina Avenue causing major delays and bunching south of King Street West, the route will no longer provide scheduled service south of Front Street West on weekdays between 3pm and 7pm. During this time, buses will loop via Front Street West and Blue Jays Way before turning back northbound. Some buses may extend to Queens Quay for customers with mobility needs or require assistance south of Front Street. Outside of these hours, all buses will continue to operate between Spadina Station and Queens Quay.
- July 30, 2024 - Afternoon rush hour service (3pm to 7pm) south of Front Street West is restored with the introduction of a new dedicated transit priority lane on Spadina Avenue between Richmond Street West and Lake Shore Boulevard West. Replacement bus service will no longer short turn at Front Street West during these hours and will instead serve all stops along Spadina Avenue.
- October 6, 2024 - Two new branches will operate to minimize traffic-related disruptions at the southern-end of the route, and to also provide replacement bus service for the 509 Harbourfront route. Branch 510C will operate to King Street West looping via Charlotte Loop, and branch 510D will operate to Exhibition Loop via Queens Quay West, Bathurst Street, Fort York Boulevard, and Fleet Street. Service will alternate between the two branches. Branch 510B to Queens Quay is suspended.
- January 5, 2025 - Branches 510C to King and 510D to Exhibition are removed. One single 510B branch will operate to Queens Quay looping via Lake Shore Boulevard West and Dan Leckie Way.

Overnight service is provided by 310 Spadina over the entire route.
