This route services Queens Quay West between Bay Street and Bathurst Street. It also provides service on Fleet Street and Manitoba Drive. Its eastern terminal is Union Station and the western terminal is Exhibition Loop for its one branch, 509.

History
- July 23, 2000 - New streetcar route is introduced operating between Union Station and Exhibition Place via Queens Quay West and Fleet Street. Most service will operate between Union Station and Exhibition except during the early mornings when streetcars only operate to Fleet Loop at Lake Shore and Bathurst. Although service is to operate year-round, more frequent service will be provided during the summer months to match the seasonal ridership changes.
- May 11, 2003 - Late evening seasonal service added every day.
- May 8, 2011 - Route moved to Russell Division because of construction at Roncesvalles.
- March 29, 2015 - Debut of the Bombardier Flexity
streetcars on the route.
- October 11, 2015 - Route is added to the Ten Minute Network. Service on this route will operate at intervals of ten minutes or better whenever the subway is open.
- January 3, 2016 - Route split at Fleet Street loop due to construction on the Exhibition loop. 509/511 replacement bus replaces service west of Fleet Street to the Exhibition.
- August 13, 2016 - With construction completed, full streetcar service is restored to Exhibition Loop.
- September 1, 2024 - All service is converted to bus operations due to overhead upgrades along Queens Quay West. Replacement bus service will operate between Exhibition Loop and Union Station. Phase 1 of the overhead upgrades will occur east of Spadina Avenue.
- October 6, 2024 - With Phase 2 of the Queens Quay West overhead upgrades which occurs between Bathurst Street and Spadina Avenue, streetcar service is partially restored operating between Union Station and Queens Quay/Spadina loop. Replacement bus service on the new 510D Spadina
branch will serve the section between Spadina Avenue and Exhibition Loop.
Overnight Service
Overnight service is provided by 310 Spadina on Queens Quay West between Bay Street and Spadina Avenue. The rest of the route does not have overnight service, however, this route operates until 1:00 AM every day.
