21 Milton is a bus route operated by GO Transit. It serves as the Train-Bus replacement for the Milton GO Train during off-peak periods, although service to Square One GO Terminal is retained daily as well. This is a daily service. It is part of the Milton series of GO Transit routes.

21 Milton is one of the busiest bus routes operated by GO, serving as a link between Downtown Toronto and many parts of Mississauga, including Square One. The route is split into at least 2 sections for the majority of the operating hours. It also previously has the most branches of any GO route, with 18 lettered branches. As of April 27th, 2024 it will have 7 lettered branches.

From April 8th, 2023-April 27th, 2024 Route 21 was realigned removing daily service to Union Station GO Bus Terminal. The 5 branches to operate 21 Milton-Union Station (All stops/Early Mornings, Late Nights) 21A Milton-Oakville, 21B Lisgar-Clarkson, 21C Erindale-Port Credit, 21D Lisgar-Clarkson (skipping Streetsville GO, and Erin Mills Transitway Station) to connect customers to the Lakeshore West GO Train which would take customers into Union Station to avoid heavy delays on the Gardiner Expressway due to long term construction. After huge customer backlash and complaints service was realigned back to Union Station GO Bus Terminal for the April 27th, 2024 Board change, All branches were re-lettered and no longer stops at Dixie GO.

Customers traveling from Milton GO Station, Derry & 9th Line, and Meadowvale Town Centre can connect at Meadowvale GO Station to Route 27B if they are traveling towards Finch Bus Terminal.
