This route services Laird Drive south of Eglinton Avenue East, Millwood Road, and Donlands Avenue. It also provides service on Eglinton Avenue East between Yonge Street and Laird Drive. There is one southern terminal, which is Donlands Station. Northern terminals include Eglinton Station for 56A and Brentcliffe Road for 56B.

History
- November 23, 2008 - Moved to shared operation between newly-opened Mount Dennis and Wilson Divisions.
- May 8, 2011 - Full operation moved to Wilson Division. Also, due to system-wide service cuts, service is removed after 10pm on weekdays and after 7pm on weekends.
- June 22, 2014 - Due to a system-wide reorganization of route branch designations, branch 56 to Eglinton Station has been changed to 56A.
- September 6, 2015 - Service after 10pm on weekdays and after 7pm on weekends is restored.
- March 29, 2020 - Due to Metrolinx Line 5 construction restricting left turns at the Yonge/Eglinton intersection, service on branch 56A is rerouted to enter Eglinton Station via south on Duplex Avenue and east on Berwick Avenue.
- October 13, 2020 - Due to the COVID-19 pandemic and the reallocation of service to other busier routes, all service on short-turn branch 56B to Brentcliffe is temporarily suspended until further notice. No service will be provided on Brentcliffe Road.
- March 28, 2021 - This route was transferred to Eglinton upon the opening of the brand new McNicoll Division.
- March 31, 2024 - Buses now revert to its pre-Line 5 construction routing and now enter Eglinton Station via south on Yonge Street and west on Berwick Avenue. This change was made in January 2024 but is now in official schedules.
Overnight Service
Overnight service is provided by 334 Eglinton East and 354 Lawrence East on Eglinton Avenue East between Yonge Street and Brentcliffe Road. The rest of the route does not have overnight service, however, this route operates until 1:00 AM every day.
