This route services Islington Avenue between Bloor Street West and Lake Shore Boulevard West. It also provides service on Judson Street, Horner Avenue, 30th Street, and Browns Line. There is one northern terminal, which is Islington Station. Southern terminals include Long Branch Loop for 110A and 110B, and Kipling Loop at Lake Shore Boulevard West for 110C. Branch 110B only operates during the rush hour on weekdays.

History
- November 23, 2008 - Service extended until 1:00am every day on branch 110A to Long Branch via Horner.
- August 2, 2009 - Route is made bike rack equipped.
- June 22, 2014 - Due to a system-wide reorganization of route branch designations, branch 110 to Lake Shore is changed to 110C.
- March 26, 2023 - Branch 110C to Lake Shore is extended west along Lake Shore Boulevard to Kipling Loop. As a result of a turn restriction from Lake Shore Boulevard, buses will no longer turn north on Twelfth Street and instead use Thirteenth Street.
Overnight Service
Overnight service is provided by 337 Islington on Islington Avenue between Bloor Street West and Lake Shore Boulevard West only. The rest of the route does not have overnight service, however, the route operates until 1:00 AM every day.
