This route services King Street between Spadina Avenue and Queen Street East. It also provides service on Queen Street East between King Street East and Kingston Road, and Kingston Road between Queen Street East and Victoria Park Avenue. Its western terminal is Spadina Avenue (Charlotte Loop) and the eastern terminal is Victoria Park Avenue (Bingham Loop) for its one branch, 503.

History
- June 21, 2015 - With service increases on 502 Downtowner and 503 Kingston Rd streetcar service during the midday on weekdays, and on branch 22A during all other times, combined service along Kingston Road west of Victoria Park Avenue will operate at intervals of ten minutes or better whenever the subway is open.
- January 4, 2016 - Due to a shortage of streetcars, buses operate the entire route temporarily.
- June 20, 2016 - Streetcar service resumes.
- October 11, 2016 - Buses temporarily replace streetcar operations.
- May 8, 2017 - Streetcar service returns. Service along Wellington Street is removed due to construction and the route is extended to Spadina Avenue. Streetcars use Charlotte Loop to turn around.
- February 19, 2018 - New midday service is introduced to complement the King Street Pilot Project.
- January 7, 2019 - Due to City construction along Wellington Street, streetcar service is replaced with buses operating from Bingham Loop to York Street looping clockwise via Church Street, Wellington Street, and York Street.
- September 3, 2019 - Due to construction at Queen Street East and Kingston Road until November 2019, daytime service on Kingston Road is consolidated on this route to provide a more reliable service.
- November 25, 2019 - With the completion of construction at Queen Street East and Kingston Road, all daytime service along Kingston road continues to be consolidated on this route on a trial basis. The western looping is changed to loop via north on York Street, west on Richmond Street West, south on University Avenue and east on King Street West.
- March 23, 2020 - Due to decreased ridership as a result of the COVID-19 pandemic, service will operate between Bingham Loop and Queen Street East/Kingston Road only.
- June 21, 2020 - Full service is restored with accessible Bombardier Flexity operating over the entire route. Service will operate between Bingham Loop and Charlotte Loop at Spadina Avenue.
- July 31, 2022 to September 4, 2022 - With track repairs at the intersection of King Street East and Sumach Street, service will divert both ways via Parliament Street and Queen Street East.
- November 20, 2022 - Due to overhead conversion along Kingston Road, the route is converted to a full bus operation.
- May 7, 2023 - Route suspended and replaced with the extended 505 Dundas streetcar route operating to Bingham Loop via Broadview Avenue, Queen Street East, and Kingston Road.
- July 4, 2023 - Route is reinstated as a non-scheduled route using "extra" buses operating until 8pm every day.
- July 30, 2023 - Route now operates as a scheduled service. Buses operate between York Street and Bingham Loop every day whenever the subway is open.
- October 8, 2023 - Streetcars resume operation of the route between Charlotte Loop (Spadina Avenue) and Bingham Loop daily.

Overnight service is provided by 301 Queen on Queen Street East, 304 King on King Street, and 322 Coxwell on Kingston Road.
