This route services Sentinel Road between Sheppard Avenue West and The Pond Road. It also provides service on The Pond Road west of Sentinel Road and on Ian MacDonald Boulevard north of Shoreham Drive. Its southern terminal is Sheppard West Station and the northern terminal is Pioneer Village Station for its one branch, 106.

Orion VII NG #1383 on route 106.
- September 3, 2000 - Vehicles leaving operational service at York University remain in service while operating southbound to Sentinel Avenue and Finch Avenue to service stops in the south part of York University.
- August 31, 2008 - Bike rack service is added to route 106.
- November 23, 2008 - Moved to Wilson Division.
- September 4, 2016 - Moved to Arrow Rd. Division.
- December 17, 2017 - With the opening of the new Line 1 extension, the northern terminus is changed from York University to Pioneer Village Station and the route is renamed from 106 York University to reflect the main thoroughfare that this route serves. Moved to Wilson Division.
