Route 53 serves the East Galt and North Galt neighbourhoods. It also connects to [[Grand River Transit Ainslie Street Terminal|Ainslie Terminal]] and [[Grand River Transit Cambridge Centre Station|Cambridge Centre Station]].
 
From Ainslie Terminal, buses operate via south on Ainslie Street, east on Concession Street, south on Christopher Drive, east on Champlain Boulevard, north on Franklin Boulevard, west on Bishop Street, and south on Hespeler Road to Cambridge Centre Station. Branch 53A follows the regular routing to Franklin and Savage, diverting via east on Savage Drive, north on Dobbie Drive, and west on Clyde Road to Franklin. Buses in the opposite direction follow the reverse routing.
