From Castle Downs Transit Centre, buses operate south on Castle Downs Road, west on 132 Avenue, south on 127 Street, south on Groat Road, west on 115 Avenue, south on 135 Street (looping through Westmount Transit Centre), east on 107 Avenue, south on Groat Road, east on 87 Avenue, north on 114 Street, and east on 89 Avenue to University Transit Centre. In the opposite direction, buses operate south on 112 Street, west on 83 Avenue, and north on 114 Street to 87 Avenue, then follow the reverse routing to Westmount Transit Centre, then north on 135 Street, east on 114 Avenue to Groat Road, and follow the reverse routing to Castle Downs Transit Centre.
Route 51 serves the neighbourhoods of Beaumaris, Dunluce, Caenarvon, Griesbach, Carlisle, Rosslyn, Kensington, Calder, Wellington, Athlone, Hagmann Estate Industrial, Sherbrooke, Prince Charles, Inglewood, Woodcroft, North Glenora, Glenora, Westmount, Windsor Park, and Garneau.
