The route leaves Downtown Vancouver from Melville Street at Thurlow and performs a clockwise loop by Thurlow and West Pender to access Burrard Street. It travels south along Burrard crossing the Burrard Street Bridge onto Cornwall Avenue, Point Grey Road and then Macdonald Street. Trips operating as 2 Macdonald–16th Avenue turn west onto W. 16th Avenue and terminate at Carnarvon by doing a U-turn on W. 16th at Trutch. Trips operating as 2 Macdonald continue south on Macdonald Street, west on King Edward, south on Quesnel Drive becoming MacKenzie, west on W. 41st Avenue to Dunbar Loop.
