Route Details
AM: The southern terminus is at Old Colony Road and Bayview Avenue. Buses travel via west on Old Colony Road, north on Yonge Street, east on Sunset Beach Road, south on Bayview Park Lane, and north on Bayview Avenue to the Snively subdivision and looping via Snively Street, Worthington Avenue, and Bloomington Road. It returns south on Bayview Avenue and travels west on North Lake Road and Lakeland Crescent to Yonge Street and operates via Ashfield Drive, Bayswater Avenue, Lorridge Street, Worthington Avenue and Yonge Street. Buses operate a double-loop via Coons Road, Bloomfield Trail, and Bloomington Road to serve Cardinal Carter High School. After completing a second loop, it then continues west on Bloomington Road, north on Bathurst Street, and east on Wellington Street West to terminate at Aurora High School.
PM: Buses begin operating from Cardinal Carter Catholic High School. Buses operate over the reverse routing, except it follows the same AM looping via Bayview Avenue, Snively Street, Worthington Avenue, and Bloomington Road. Also, buses follow the same AM looping via Old Colony Road, Yonge Street, and Sunset Beach Road to terminate at Bayview Park Lane.
History
- September 7, 2010 - Route 429 started service during the 2010 school season on September 7, 2010 as a replacement for route 443, and to serve students going to Aurora High School. It largely follows the same route today, except it went directly to Bloomington Road serving the school from Yonge Street.
- February 24, 2013 - To accommodate the elimination of route 84A, the route was modified to serve Coons Road and Bloomfield Trail and buses use the loop within the school instead of stopping on-street. Also, due to the redevelopment of Lake Wilcox Park requiring the permanent closure of Olde Bayview Avenue, service between North Lake Road and Sunset Beach Road is instead rerouted via Bayview Avenue and Bayview Park Lane.
- November 1, 2015 - At Cardinal Carter High School, buses no longer use the loop within the school property. Only the morning trip loops via Yonge Street, Coons Road, and Bloomfield Trail. The afternoon trip instead operates direct via east on Bloomington Road and Yonge Street to route. In addition, the route is extended eastward along Old Colony Road from Yonge Street to Bayview Avenue. School trips now start and end at Bayview Avenue and Old Colony Road.
- September 3, 2019 - The afternoon southbound routing is modified to operate via south on Bayview Avenue, west on Old Colony Road, north on Yonge Street, and east on Sunset Beach Road to terminate at Bayview Park Lane. Afternoon trips will no longer operate west along Sunset Beach Road.

- September 2003 - Cardinal Carter was one of the first high schools to have a school special. The route was named "Cardinal Carter School Special #1" and was operated as a portion of route 84.
- September 5, 2004 - Originally unnumbered, the route is assigned the number 443.
- September 4, 2005 - The route was temporarily discontinued to start the 2005 school season.
- January 2, 2006 - Route was reinstated.
- April 26, 2009 - The southern-end of the route is modified to operate both ways via Bernard Avenue, Yorkland Street, Brookwood Drive, Devonsleigh Boulevard, and 19th Avenue to Yonge Street. Buses no longer use Yonge Street between 19th Avenue and Bernard Avenue.
- January 4, 2010 - The southern terminus is changed to Yonge Street at Old Colony Road and buses no longer operate to Bernard Terminal.
- September 5, 2010 - The route was discontinued permanently and the routing is partially replaced with the new route 429.
