This route services Lawrence Avenue East between Leslie Street and Starspray Boulevard. It also provides service on Eglinton Avenue East between Yonge Street and Leslie Street, and Leslie Street between Lawrence Avenue East and Eglinton Avenue East. Western terminals include Eglinton Station for 54A and 54B, and Don Mills Road for 54D. Eastern terminals include Starspray Boulevard for 54A, Orton Park for 54B, and Morningside Avenue for 54D. Branch 54B operates north via Scarborough Golf Club Road before 3:00pm and north via Orton Park Road after 3:00pm.

History
- June 11, 2000 - Two express stops are added on the 54E branch at Rushley Drive and Midland Avenue. Buses began serving the stops in February 2000 but this service modification was made official on this date.
- November 23, 2008 - Full operation transferred to Eglinton Division.
- June 22, 2014 - Due to a system-wide reorganization of route branch designations, branch 54 to Orton Park is changed to 54B.
- October 11, 2015 - Route is added to the Ten Minute Network. Combined service west of Orton Park Road will operate at intervals of ten minutes or better whenever the subway is open.
- September 5, 2017 - New express stop added eastbound at Midland Avenue on express branch 54E to align with the westbound stop.
- August 31, 2018 - Last day of service for the express 54E branch. As part of the TTC's initiative to reorganizing surface express bus routes, this branch will be replaced with 954 Lawrence East Express
.
- May 7, 2023 - New tripper service introduced on weekend daytime and early evenings operating between Don Mills Road and Morningside Avenue. Three buses are assigned to this service and will be designated as branch 54D. In the west, buses loop using Don Mills Road and The Donway West, and in the east, buses loop using Kingston Road and Collinsgrove Road.
- October 8, 2023 - Branch 54D (Don Mills-Morningside) is expanded to operate scheduled service every 30 minutes during the weekday midday periods.

Overnight service is provided by 354 Lawrence East along the entire 54A branch except on Leslie Street between Eglinton Avenue East and Lawrence Avenue East and Lawrence Avenue East between Leslie Street and Don Mills Road.
