This route services Dundas Street West from Bloor Street West to Kipling Avenue, except between Mabelle Avenue and Cordova Avenue. This route also provides service on Jane Street between Dundas Street West and St Clair Avenue West, St Clair Avenue West between Jane Street and Runnymede Road, and on Runnymede Road between Dundas Street West and St Clair Avenue West. Its eastern terminal is Dundas West Station. Western terminals include Kipling Station for 40A and Jane Street for 40B.

History
- January 2, 2005 - Route made accessible with low-floor buses.
- November 23, 2008 - Moved to newly-opened Mount Dennis Division.
- September 2, 2012 - Route reassigned back to Queensway Division.
- October 13, 2019 - As part of the Junction Area Study, service is extended over the former 30 Lambton routing along Dundas Street West to Kipling Station. Route is renamed from 40 Junction to 40 Junction-Dundas West to reflect this service change.
- March 30, 2020 - Due to COVID-19 related service reductions, branch 40B was temporarily discontinued on weekdays.
- October 8, 2023 - Branch 40B is interlined with route 49 Bloor West.

Overnight service is provided by 312 St Clair-Junction on Dundas Street West between Bloor Street West and Jane Street. The rest of the route does not have overnight service, however, this route operates until 1:00 AM every day.
