This route provides service on Dufferin Street south of Wilson Avenue. There is one northern terminal which is Wilson Station for both branches, 29A and 29C. Southern terminals include Dufferin Gate for 29A, and Princes' Gate for 29C within the CNE grounds. Branch 29C does not operate during the summer as the Exhibition grounds are often closed to special events during this time. Outside the summer months, branch 29C only operates during the rush hour on weekdays and during the day and early evenings on weekends.

- July 1, 1954 - Route established.
- June 19, 2005 - This route is made part of a 1-year bike rack pilot project.
- May 13, 2007 - A service trial on Branch 29D extends service to the Exhibition Loop.
- November 18, 2010 - The underpass at Dufferin Street and Queen Street West is opened and buses now travel directly on Dufferin Street. Service along Peel and Gladstone Avenues is removed.
- March 12, 2014 - Articulated bus service begins on this route.
- March 30, 2014 - Branches renamed to reduce confusion for customers.
- June 22, 2014 - Due to a system-wide reorganization of route branch designations, northbound branch 29A to Tycos Drive is changed to 29B and southbound branch 29D to Princes' Gate is changed to 29C. Short turn southbound branch 29C to Dufferin Station is eliminated.
- March 29, 2015 - Southbound branch 29 to Dufferin Gate is changed to 29A.
- June 21, 2015 - Route is added to the Ten Minute Network. Service now operates at intervals of ten minutes or better whenever the subway is open.
- October 7, 2018 - Operation of this route is transferred to Mount Dennis division. Peak-period branch 29B to Tycos Drive is eliminated with all service operating between Wilson Station and the Exhibition. Express branch 29E between Dufferin Station and Exhibition is discontinued with the introduction of the new 929 Dufferin Express route.
- January 3, 2021 - All service is temporarily extended to Princes' Gate Loop due to service alterations from the King-Queen-Queensway-Roncesvalles (KQQR) construction causing increased traffic within the loop at Dufferin Gates.
- October 8, 2023 - Due to long-term construction at Dufferin Gates Loop, service will divert to Princes' Gates Loop via Liberty Street, East Liberty Street, Strachan Avenue, Manitoba Drive, and Canada Boulevard. Buses will use the new 29D branch designation.
- January 7, 2024 - With the completion of construction within Dufferin Gates' Loop, buses return to using Dufferin Gates Loop as the southern terminus. Branch 29D is discontinued.

Express service on Dufferin Street is provided by 929 Dufferin Express between Wilson Station and Dufferin Gate Loop.
