Route 5 serves the southern portion of Milton, specifically the Timberlea and Coates neighbourhoods.
From Milton GO Station, buses operate east on Drew Centre, south on Thompson Road, west on Childs Drive, south on Ontario Street, east on Laurier Avenue, and south on Holly Avenue, looping via east on Yates Drive, south on Philbrook Drive, west on Hepburn Road, and north and east on Yates Road to Holly Avenue; then return via the reverse routing to Milton GO.

Service Summary
Route 5 operates 30 minute headways during AM and PM rush hour, and 60 minute headways during weekday midday and Saturday service. Buses used are conventional 40 foot vehicles, with Arboc Spirit of Freedom cutaway buses occasionally used as the second bus during rush hours.
Buses interline with route 8 Willmott at Milton GO.
