From Conestoga College, buses operate via west on Doon Valley Drive, south on Conestoga College Boulevard, northwest on Homer Watson Boulevard, west and north on Old Carriage Drive, west on Pioneer Drive, west on Bechtel Drive, north on Pioneer Drive, west on Black Walnut Drive, north on Biehn Drive, west on Old Huron Road, north on Battler Road, west on Huron Road, north on Strasburg Road, northeast on Ottawa Street, west on Hoffman Street, northwest on Highland Road, northwest on Belmont Avenue, north on Union Drive, and north on King Street to Waterloo Public Square Station, looping via north on King Street, west and south on Caroline Street, east on William Street, south on King Street, south on Mt Hope Street, and west on Park Street to Union Boulevard, then following the reverse routing back to Conestoga College.

Route 16 connects to Ion at Waterloo Public Square Station, Willis Way Station, and Grand River Hospital Station.
