Route 401 serves Simcoe St, Wentworth St, and Lakeview Park Dr.
This route serves the Durham College/Ontario Tech University North Campus, Downtown Oshawa, and Lakeview Park via the Simcoe/Centre Street Corridor. A short turn branch operates from the North Campus to Downtown, and a rush hour extension serves the Oshawa GO Station
Route 401 connects to 900 Pulse in Downtown Oshawa.

History
Route 401 was operating along its primary arrangement when DRT inherited the Oshawa Transit Commission in 2006. OTC denoted the #1 to the route. Slight modifications were made to the route in 2016 to have the main branch service First Street, McNaughton Avenue and Ritson Road to and from Lakeview Park.
In 2018 service was reverted back to the original alignment of route 401, route 401B continued service on Ritson to Wentworth.
In 2019, service on route 401C to Oshawa Station was removed and rebranded as new route 422 Simcoe/Bloor which mirrors the 401 from Simcoe/Commencement to Simcoe/Centre and Gibb, before servicing Park Road, and Bloor Street to Oshawa Station.

Route 401 only uses 40 foot vehicles with service provided by Nova Bus LFS, New Flyer D40LF or New Flyer XD40.
