The earliest precursor to this route was the former 203 Centennial GO Shuttle, which operated along a similar routing. It was not successful and was discontinued.
Plans for route 45 have existed for many years, always appearing on maps for proposed service changes, but never being implemented. Mingay was approved to be implemented in 2008, but due to limited vehicle resources implementation of this initiative was deferred pending vehicle availability.
- September 8, 2009 - Route 45 begins service.
- December 2012 - Route 45 was declared fully accessible.
- April 2020 - Service on route 45 was temporarily suspended in response to ridership changes due to COVID-19.
