The earliest precursor to this route was the former 203 Centennial GO Shuttle, which operated along a similar routing. It was not successful and was discontinued.
Plans for route 45 have existed for many years, always appearing on maps for proposed service changes, but never being implemented. Mingay was approved to be implemented in 2008, but due to limited vehicle resources implementation of this initiative was deferred pending vehicle availability. It was implemented on September 6, 2009, with the first run on September 8.
Route 45 was declared fully accessible in December 2012.
