Route 16 mainly serves 16th Avenue and Carville Road between Bathurst Street and Ninth Line.
This route serves the Patterson and Cornell communities. It also serves St. Brother Andre Catholic High School and Cornell Community Centre.
Route 16 connects to Viva Blue at Yonge Street and Viva Purple at Cornell Terminal.

History
- 2013 - Route 16 was created from the 2013 Annual Service Plan, which restructured the Rutherford Road/16th Avenue corridor in order to increase reliability of service. The east half of route 85 was renumbered to this current route. Route 85A became the new 85 Rutherford, providing service between Napa Valley Avenue and Yonge Street, with service operated by Southwest Division. Route 85B was discontinued, and service in the Vellore Woods community was replaced by new route 21. Service in the Beaver Creek Business Park was replaced by the reinstated route 244.
- July 28, 2013 - Routing was adjusted to operate via Bathurst Street and Lebovic Campus Drive on westbound trips, and Lebovic Campus Drive, Ilan Ramon Boulevard, Rutherford Road, and Markham Stouffville Hospital on eastbound trips.
- September 4, 2022 - With the opening of the Cornell Terminal, routing is adjusted to serve the terminal as the new eastern terminus.
