General
Route 46 operates between Erin Mills Transitway Station and Meadowvale Town Centre Bus Terminal. It also serves Erin Mills Town Centre Bus Terminal.
The route primarily serves Tenth Line, Osprey Boulevard and Trelawny Circle. It provides service to the Central Erin Mills, Churchill Meadows, Lisgar, and Meadowvale neighborhoods of Mississauga.

Timeline
- September 2, 2008 - Route 46 'Tenth Line' is introduced, providing service between Erin Mills Town Centre and Lisgar GO Station via Erin Centre Boulevard and Tenth Line. Service is provided during weekday rush hour periods with 30 minutes frequency.
- July 2, 2012 - Route designated as accessible, with low floor buses used at all times.
- October 24, 2016 - Route renamed 'Tenth Line-Osprey' from 'Tenth Line'. Routing in the south-end extended to serve Erin Mills Transitway Station. Routing north of Britannia Road is revised to use Grossbeak Drive, Osprey Boulevard, Trelawny Circle, and Aquitaine Avenue to Meadowvale Town Centre, no longer serving Lisgar GO Station. Service is provided 7 days a week with weekday peak frequency 20 minutes; weekday midday frequency 23 minutes; weekday evening frequency 35 minutes; Saturday frequency is 23 minutes; Sunday frequency is 35 minutes. Route interlines with route 48 on weekdays.
- July 1, 2024 - As part of a MiWay initiative to simplify the route naming convention, the route is renamed 'Tenth Line' from 'Tenth Line-Osprey'.
