Route 31 provides service between Guelph and Union Station Bus Terminal, serving all stations on the Kitchener line outside of Toronto between Guelph Central GO and Malton GO. The route also provides service along Highway 7 between Guelph and Brampton in the communities of Rockwood, Acton, and Georgetown. It serves the University of Guelph, Shopper's World, and Brampton Bus Terminal.

Service operates both as a replacement of the Kitchener line when trains do not operate, and as a train-meet service with the Kitchener line. Trips on branches 31B and 31R are scheduled to connect with trains at Bramalea GO.

Service is primarily provided in the counter-peak direction on weekdays, and also during early morning and late evening periods. Service west of Shopper's World is primarily provided on route 33 during the daytime. Some additional service is provided on the 31M branch during fall and winter months to accommodate demand to the University of Guelph.
