Route 603 Pickering Port Perry, begins its five daily northbound trips at Glenanna and The Esplinade, then travels in a loop around Pickering Parkway Terminal and the Pickering Town Centre via Glenanna, Pickering Parkway and Liverpool, the route then travels east on Kingston Road to Brock Road (R.R. 1), Route 603 travel north on R.R. 1 serving the GO Transit Brock Road and Highway 407 Park and Ride, serving the communities of Brougham and Claremont. the route travels northeast on Highway 47 into Uxbridge servicing the terminal at 6 Welwood Drive where two trips terminate. The remaining three trips continue to the Downtown Transfer Point at Toronto and Brock St before traveling east on Reach Street into Port Perry.
Southbound trips utilize Brock Road to Pickering Parkway, then to the Pickering Parkway Terminal, Liverpool Road, Kingston Road, and Glenanna to the terminus. Only two southbound trips begin in Port Perry.
