It operates along Regional Road 1 from Uxbridge through Leaksdale and Udora, then on Regional Road 10 through Sunderland, on Highway 7 through Manilla, on Simcoe Street through Cannington, on Regional Road 12 through Wilfrid, on Regional Road 23 to Beaverton. Connection to GO Transit is available at various points. A connection to YRT route 50 is available at Highway 48.
The route was launched as the Brock Community Bus and operated mostly on Highway 47 and Highway 12. In May 2015, the route was given the number 601 and more trips were added. The route was adjusted in 2016 to run on Regional Road 1 through Leaksdale and Udora, on Highway 7 through Manilla, and Regional Road 12 and Regional Road 23 through Wilfrid.
