The 501 originated as Pre-DRT route 1 Aspen Springs until December 31st, 2005. The 1 Aspen Springs was given the 50x prefix in 2008 when DRT was simplifying their route numbering system. On June 27th 2016 the route 501 Aspen Springs was renamed and rerouted to become route 501 Bowmanville South as we know it today.
