The 406 ran south, originating from King and Centre Streets, Downtown Oshawa. The route utilizes side streets such as Dean, Shakespeare, Tennyson and Farewell. It returns north via Ritson Road South and McNaughton Avenue. The 406 connects passengers from the south east end of Oshawa, to Downtown. The route interlines with 407/B at Simcoe and Bond Streets.

The 406 Dean dates back to Oshawa Transit era, since this time, no major changes have occurred. The route originally carried a #6 until a #4 was added to all Oshawa Routes in 2008. The route was canceled in 2015 when the 407 Ritson was extended and absorbed the full route.
