This route services Cummer Avenue and McNicoll Avenue between Yonge Street and Dynamic Drive. There is one western terminal, which is Finch Station and there is one eastern terminal, which is Dynamic Drive for its one branch, 42A.

History
- September 7, 2004 - Weekday midday service added on branch 42A.
- October 14, 2005 - Weekday midday service on branch 42A is discontinued.
- January 14, 2007 - All service on 42B branch is extended to serve a new McNicoll/Kennedy Loop east of Kennedy Road.
- November 23, 2008 - Moved to Wilson Division. Off-peak service added east of Kennedy Road and extended until 1:00am on the 42A branch.
- May 9, 2011 - Due to system-wide cuts, service is removed on the 42A branch after 10pm on weekdays, and all day on weekends and holidays.
- June 22, 2014 - Branch 42 eastbound to Victoria Park is redesignated 42C due to a system-wide reorganization of route branch designations.
- September 6, 2015 - Service on the 42A branch after 10pm on weekdays, and all day on weekends and holidays except late evenings is restored.
- January 3, 2016 - Branch 42A is extended east to Dynamic Drive to provide service to the Tapscott Employment District.
- June 19, 2017 - The off-street loop east of Kennedy Road used by branch 42B will be removed as part of the construction of the TTC's new McNicoll Garage. All service on the 42B branch will operate as 42A.
- October 13, 2020 - Due to the COVID-19 pandemic, branch 42C to Victoria Park is suspended until further notice to reallocate resources to busier routes.
- March 28, 2021 - This route was transferred to the brand new McNicoll Division.
- February 14, 2022 - Service on branch 42C is resumed.
- June 18, 2023 - Peak-period branch 42C to Victoria Park is discontinued. Service at all times will operate to Middlefield Road and Dynamic Drive.
Overnight Service
Overnight service is not provided, however, the route operates until 1:00 AM every day.
